He is best known for his role as Sergeant Warren Reed in the Robocop films, as well as his roles in Coffy, Treasure of Matecumbe, Guyana: Crime of the Century, and Miracle Mile. He also appeared in three films by Robert Altman and provided the voice of Pablo Robertson in the animated Harlem Globe Trotters TV series. DoQui also had guest spots on many TV shows, including NYPD Blue, Star Trek: Deep Space Nine, Picket Fences, and The Jeffersons. He served ten years on the board of directors of the Screen Actors Guild, encouraging women and minority groups to participate more in the media.

He starred in the miniseries Centennial in 1978, and the television film The Court-Martial of Jackie Robinson in 1990. He made guest appearances on many television series, including I Dream of Jeannie, Happy Days, The Jeffersons, Daniel Boone, Gunsmoke, Adam-12, The Parkers, and Star Trek: Deep Space Nine in the season 4 episode "Sons of Mogh" as a Klingon named Noggra.
